A Data-Driven, Mathematical Model of Mammalian Cell Cycle Regulation.This article is from PLoS ONE , volume 9 . Abstract Few of >150 published cell cycle modeling efforts use significant levels of data for tuning and validation. This reflects the difficultly to generate correlated quantitative data, and it points out a critical uncertainty in modeling efforts. To develop a data-driven model of cell cycle regulation, we used contiguous, dynamic measurements over two time scales (minutes and hours) calculated from static multiparametric cytometry data. The approach provided expression profiles of cyclin A2, cyclin B1, and phospho-S10-histone H3. The model was built by integrating and modifying two previously published models such that the model outputs for cyclins A and B fit cyclin expression measurements and the activation of B cyclin/Cdk1 coincided with phosphorylation of histone H3. Abstract The proliferation of vascular smooth muscle cells may perform a crucial role in the pathogenesis of diabetic vascular disease. AMPK additionally exerts several salutary effects on vascular function and improves vascular abnormalities. The current study sought to determine whether sodium tanshinone IIA silate (STS) has an inhibitory effect on vascular smooth muscle cell (VSMC) proliferation and migration under high glucose conditions mimicking diabetes without dyslipidemia, and establish the underlying mechanism. In this study, STS promoted the phosphorylation of AMP-activated protein kinase (AMPK) at T172 in VSMCs. VSMC proliferation was enhanced under high glucose (25 mM glucose, HG) versus normal glucose conditions (5.5 mM glucose, NG), and this increase was inhibited significantly by STS treatment. Methods: PaTu8988 pancreatic cancer cells were treated with different concentrations of suberoylanilide hydroxamic acid (SAHA), cell survival, proliferation, migration and vasculogenic mimicry (VM) were analyzed. Associated signaling changes were also analyzed by RT-PCR and Western blots. Results: Here, we reported that SAHA, a histone deacetylase inhibitor (HDACi), exerted significant inhibitory efficiency against pancreatic cancer cell survival, proliferation, migration and VM. DTIC ADA316821: Cellular Proteins Interacting with the Tumor Suppressor Protein p53.Tumor suppressor protein p53 interacts directly with the DNA replication factor RPA and inhibits its ability to bind single-strand-DNA. We defined the domain of p53 that bound to RPA and constructed p53 mutants that failed to bind RPA, but still functioned as transcriptional activators. We found that while these mutants of p53 lost their ability to bind RPA; they still maintained the growth suppression function of p53. Growth suppression function of p53 is dependent on its transactivation activity, probably by inducing p21 and other cell cycle inhibitors. We have extended our study to the p2l protein, which is induced by p53 and interacts with both the cdk2 Icinase and a DNA replication factor PcNA. Here we have demonstrated the importance of PCNA-inhibitory domain of p21 in vivo. Coordination of these events is essential. I have used the budding yeast S. cerevisiae to dissect the molecular events that control cell cycle progression. I have shown that not only are G1 cyclin-CDK complexes highly unstable rate-limiting activators for cell cycle progression, but that their essential function is to regulate the initiation of DNA replication by phosphorylating and promoting the degradation of a key cell cycle inhibitor, Sic1 (1,2). In fact, cyclin-CDK-regulated proteolysis may constitute the major mechanism of cell cycle control (3). Moreover, Sic1 appears to be the analog of the RB tumor suppressor gene. Either the loss of p27 or over-expression of its key regulator ubiquitin E3 ligase SKP2 is associated with prostate cancer. Prostate-sepcific expression of SKP2 is sufficient to induce hyperplasia, displasia, and low grade carcinoma in the mouse prostate gland. We propose to examine whether SKP2 cooperates with or is regulated by tumor suppressors such as Pten or Nkx3.1 implicated in prostatic tumorigenesis. In the past year, we have initiated the experiments by breeding and expansion of the colonies, crossing the SKP2 transgenic mice into Pten +/- strains, and ordering Nkx3.1 mice. Cancer is essentially a disease of inappropriate proliferation due to a loss of normal cell cycle controls. We study cell cycle controls in the budding yeast S. cerevisiae. We have shown that the formation of active G1-cyclin dependent kinase complexes (Cdks) is rate limiting for cell cycle progression. The activity of these complexes is regulated through their association with highly unstable G1 cyclins. G1 cyclins are constitutively unstable, and their only essential function is to phosphorylate the B-type Cdk inhibitor, Sic1, and target it for degradation. Sic1 is one of few known in vivo substrates of yeast G1-Cdks. The amount and phosphorylation state of Sic1 appears to determine the timing and size of cell division. Here, a new approach is built on previous methodologies in which high-throughput data was interpreted using prior biological knowledge of cause and effect relationships.
