| DTIC ADA386852: Role of cdc25 Phosphatases in Cellular Immortalization | The goal of this proposal is to study the potential role of tyrosine phosphorylation-dependent checkpoint and especially the cdc25 phosphatase family of cyclin dependent kinase (CDK) activators in cellular immortalization. We have shown that cdc25 phosphatases did in fact extend the normal mammary epithelial cells (HMEC) life span, but do not immortalize them. The effect was most prominent with cdc25A and cdc25B phosphatases. Cdc25C phosphatase had less effect on HMEC life span. Experiments with catalytically inactive cdc25A mutant show that phosphatase activity of cdc25A is essential for the life span extension. Our experiments show that telomerase is not activated by cdc25A expression in normal human cells, suggesting that cdc25 most likely affect Ml, but not M2 checkpoints. Akt, a downstream kinase effector of PI3K, stabilizes cyclin Dl protein and phosphorylates the cdk inhibitor p21 preventing its anti-proliferative effects in breast cancer cells. In this report we attempt to identify PI3K- dependent molecules necessary to rescue cyclin A expression. We examined if loss of p27 together with forced expression of cyclin Dl was sufficient to rescue cyclin A expression in p27-null MEFs and demonstrated that S phase entry could not be restored by forced cyclin Dl expression. | DTIC ADB265310: Role of Cyclin D1 and cdk Inhibitors in Breast Cancer Pathogenesis | Extensive molecular, biochemical, cell biological, and cytogenetic studies of breast cancer cell lines and breast cancer biopsies have indicated that overexpression of cyclin Dl plays an important role in breast tumor oncogenesis. Particularly, D-type cyclins are fundamental to cell cycle regulation of the tumor suppressor protein, Rb. | DTIC ADA416606: Mapping Critical DNA Sequence Elements Required for Amplification of erbB2 in Breast Cancer | To study the implications of replication initiator proteins to gene amplification, we characterized two proteins required for DNA replication (ORC and Mcm10). In an attempt to purify recombinant human ORC from insect cells infected with baculoviruses expressing HsORC subunits, we found ORC2, -3, -4, and -5 forms a core complex. Expression of ORC3N, which inhibits ORC2-ORC3 interaction, causes cell cycle arrest. Consistent with this result, ORC2-depletion by RNAi arrested cell cycle in G1 with low CDK activity. We propose that cells have a novel mechanism to ensure that CDK2 is activated only after enough pre-RC is formed. We also report that chromatin-loading of Mcm10 requires a functional pre-RC but is independent of Cdc7 activity. Mcm10 is required for chromatin-binding of Cdc45 and origin unwinding. Genistein has been shown to exert chemopreventive and antiproliferative in animal models of cancer and breast cancer cells respectively. Accumulating evidence suggest that genistein is oxidatively metabolized by the cytochrome P45O (CYP45O) family of enzymes, and that its metabolism is specific to breast cancer cells but not nontransformed breast epithelial cells. We investigated the cellular association and effects of metabolism of genistein by CYP45O in T47D breast cancer cells and MCFlOA breast epithelial cells. These studies show that genistein exerts distinct effects on cell cycle progression that is due to its preferential cellular uptake and extent of its metabolism in T47D cells but not MCFlOA cells.
