Anti-Cancer Effect of IN-2001 in MDA-MB-231 Human Breast Cancer.This article is from Biomolecules & Therapeutics , volume 20 . Abstract In recent years, inhibition of HDACs has emerged as a potential strategy to reverse aberrant epigenetic changes associated with cancer, and several classes of HDAC inhibitors have been found to have potent and specific anticancer activities in preclinical studies. But their precise mechanism of action has not been elucidated. In this study, a novel synthetic inhibitor of HDAC, 3-(4-dimethylamino phenyl)-N-hydroxy-2-propenamide [IN-2001] was examined for its antitumor activity and the underlying molecular mechanisms of any such activity on human breast cancer cell lines. IN-2001 effectively inhibited cellular HDAC activity (IC50 = 0.585 nM) in MDA-MB-231 human breast cancer cells. IN-2001 caused a significant dose-dependent inhibition of cell proliferation in estrogen receptor (ER) negative MDA-MB-231 human breast cancer cells. The bpVs cause phase-specific cell cycle arrest (G1/S), dose-dependent inhibition of Cdk activity, and persistent Rb hypophosphorylation upon release from serum starvation, consistent with Cdc25A inhibition. Further, they cause p53-independent apoptosis. Oxidative stress and DNA damage do not appear to be involved in their mechanism of action, given that: p53 is not induced by bpV(Me2Phen); p53 and p21 status do not affect IC50; depletion of glutathione or supplementation with antioxidants does not affect IC50, in contrast to other heavy metal-based agents. Mammary tumors, mostly carcinoma in situ, developed spontaneously in the MMTV-DNIIR mice with a long median latency (27.5 months). Invading carcinoma cells in MMTV-DNIIR animals showed loss of DNIIR transgene expression as determined by in situ hybridization. The data indicate that signaling from endogenous TGF-Betas not only plays an important role in normal mammary gland physiology but also can also suppress the early stage of tumor formation and contribute to tumor invasion once carcinomas have developed. The ability of the TGF-beta signaling pathways to inhibit proliferation of many epithelial cells while stimulating proliferation fibroblasts remains a conundrum. Abstract Background: High-throughput measurement technologies produce data sets that have the potential to elucidate the biological impact of disease, drug treatment, and environmental agents on humans. The scientific community faces an ongoing challenge in the analysis of these rich data sources to more accurately characterize biological processes that have been perturbed at the mechanistic level. Here, a new approach is built on previous methodologies in which high-throughput data was interpreted using prior biological knowledge of cause and effect relationships. These relationships are structured into network models that describe specific biological processes, such as inflammatory signaling or cell cycle progression. This enables quantitative assessment of network perturbation in response to a given stimulus.
