The arginine methyltransferase PRMT6 regulates cell proliferation and senescence through transcriptional repression of tumor suppressor genes.This article is from Nucleic Acids Research , volume 40 . Abstract The protein arginine methyltransferase 6 (PRMT6) is a coregulator of gene expression and executes its repressing as well as activating function by asymmetric dimethylation of histone H3 at R2 (H3 R2me2a). Given that elevated expression levels of PRMT6 have been reported in various cancer types, we explore here its role in cell proliferation and senescence. We find that knockdown of PRMT6 results in proliferation defects of transformed as well as non-transformed cells, causes G1-phase arrest and induces senescence. This phenotype is accompanied by transcriptional upregulation of important cell cycle regulators, most prominently the cyclin-dependent kinase (CDK) inhibitor gene p21 (p21CIP1/WAF1, CDKN1A) and p16 (p16INK4A, CDKN2A). Chromatin immuno-precipitation analysis reveals that the p21 gene is a direct target of PRMT6 and the corresponding histone mark H3 R2me2a. Abstract Background: Pancreatic cancer is one of the most aggressive human malignancies with a extremely low 5-year survival rate. Hence, the search for more effective anti-pancreatic cancer agents is urgent. Methods: PaTu8988 pancreatic cancer cells were treated with different concentrations of suberoylanilide hydroxamic acid (SAHA), cell survival, proliferation, migration and vasculogenic mimicry (VM) were analyzed. Associated signaling changes were also analyzed by RT-PCR and Western blots. Results: Here, we reported that SAHA, a histone deacetylase inhibitor (HDACi), exerted significant inhibitory efficiency against pancreatic cancer cell survival, proliferation, migration and VM. DTIC ADA372217: Cyclin D1, Anchorage-Independent Growth and Breast Cancer.Derangements in the cell cycle machinery contribute to uncontrolled cell growth and tumorigenesis. Regulators of GI progression including cyclin D1 and the cyclin-dependent kinase inhibitors p21 and p27 appear to be of particular importance in the pathogenesis of breast cancer. Our data show that the overexpression of cyclin D1 leads to a compensatory increase in the cdk inhibitor p21, and suggest that this increase can weaken the value of cyclin D1 overexpression as a diagnostic indicator. We suggest that aggressive breast cancer will involve both (1) overexpression of cyclin D1 and (2) the failure to undergo a compensatory upregulation of cdk inhibitors. The specific aims are designed to test these hypotheses in cell culture models, nude mice, and breast cancer biopsies. DTIC ADA443059: Suppression of Prostate Cancer by PTEN and p18(INK4C)The Rb pathway suppresses tumorigenesis by constraining G1 cell cycle progression. Functional inactivation or reduction of this pathway is a common event found in many types of human tumors. To test how the Rb pathway interacts with other cellular pathways in tumor suppression, we characterized mice with combined mutations in the CDK inhibitor p18 (sub Ink4c) and the lipid phosphatase Pten, which regulates cell growth and survival. Thep18-/-;Pten +/-double mutant mice develop prostate cancer in the anterior and dorsolateral lobes and thyroid C cell tumors with nearly complete penetrance, and pituitary tumors in both the anterior and intermediate lobes. AKT/PKB, an oncoprotein and downstream substrate of PTEN, was activated and accumulated at the plasma membrane in Pten+i. cells, and further activated and accumulated in the nucleus in p18-/-;Pten+/- tumor cells, suggesting a negative regulation of AKT by the Rb pathway. DTIC ADA474451: Mechanism of Antineoplastic Action of Bisperoxovanadium CompoundThis work describes studies into the mechanism of action of bisperoxovanadium (bpVs) compounds and into the basic biology and regulation of Cdc25A phosphatase. The bpVs cause phase-specific cell cycle arrest (G1/S), dose-dependent inhibition of Cdk activity, and persistent Rb hypophosphorylation upon release from serum starvation, consistent with Cdc25A inhibition. Further, they cause p53-independent apoptosis. Oxidative stress and DNA damage do not appear to be involved in their mechanism of action, given that: p53 is not induced by bpV(Me2Phen); p53 and p21 status do not affect IC50; depletion of glutathione or supplementation with antioxidants does not affect IC50, in contrast to other heavy metal-based agents. With respect to regulation, the Cdc25A nuclear localization signal (NLS) was identified and characterized. Novel structural features of CDK inhibition revealed by an ab initio computational method combined with dynamic simulationsThe rational development of specific inhibitors for the ~500 protein kinases encoded in the human genome is impeded by a poor understanding of the structural basis for the activity and selectivity of small molecules that compete for ATP binding. Combining classical dynamic simulations with a novel ab initio computational approach linear-scalable to molecular interactions involving thousands of atoms, we have investigated the binding of five distinct inhibitors to the cyclin-dependent kinase CDK2. We report here that polarization and dynamic hydrogen bonding effects, so far undetected by crystallography, affect both their activity and selectivity. DTIC ADA385811: The Regulation of Human Cyclin E Protein Levels by the Ubiquitin Proteolytic PathwayCyclin E is an unstable protein that is degraded in a ubiquitin- and proteasome-dependent pathway. Two factors stimulate cyclin E ubiquitination in vivo: when it is free of its CDK partner, and when it is phosphorylated on threonine 380. We pursued the first of these pathways by using a two-hybrid screen to identify proteins that could bind only to free cyclin E. This resulted in the isolation of human Cul-3, a member of the cullin family of E3 ubiquitin-protein ligases. We found that Cul-3 was bound to cyclin E but not to cyclin E-Cdk2 complexes in mammalian cells, and that overexpression of Cul-3 increased ubiquitination of cyclin E but not other cyclins. Conversely, deletion of the Cul-3 gene in mice caused increased accumulation of cyclin E protein, and had cell-type specific effects on S phase regulation. In the extraembryonic ectoderm, where cells undergo a standard mitotic cycle, there was a greatly increased number of cells in S phase.
