The target p16 is a tumor suppressor and a CDK inhibitor and is inactivated by gene methylation. The drug 5-aza-2'deoxycytidine (5-Aza-CdR), an inhibitor of DNA methylation, is targeted for reexpression of a repressed p16 in late stage breast cancer patients. Re-activation of the p16 gene in cells in which the gene is methylated will restore normal growth control and be efficacious in treatment of breast cancer. Many problems in biology and medicine center around cell division, but without a knowledge of the mechanism by which division is controlled, these problems are difficult to attack. The major transitions of the cell cycle are is controlled by a protein called a cyclin dependent kinase, or Cdk. The cell cycles of both yeast and human cells are controlled by a Cdk. Using the yeast two hybrid system I have isolated many novel Cdk interacting proteins in yeast, and these may be instrumental in controlling cell cycle progression. I have also isolate two exciting proteins which can only interact with a substrate trapping mutant of a yeast Cdk. These may be Cdk new substrates. I have also used the genomic technology of Gene Chips, to define the complete set of cell cycle regulated genes in yeast, and to characterize their controlling promoter elements. DTIC ADA411262: Targeting the AP-1 Transcription Factor for the Treatment of Breast CancerThe AP-l transcription factor is a central component of many signal transduction pathways. We have shown that blocking AP-l by over-expressing a dominant negative form - of cJun (cjun-DN, Tam67) inhibits breast cancer cell growth. We hypothesize that- inhibition of AP-l blocks the cell cycle, reverses TAM-resistance of breast cancer cells, and suppression of AP-l in vivo causes regression of existing breast tumors. In the present study, we demonstrated that TAM67 inhibits breast cancer growth both in vitro and in vivo. We determined the mechanism by which AP-l blockade inhibits breast cancer growth. Our studies suggested that TAM67 inhibits breast cancer growth predominantly by inducing CDK inhibitors (such as P27), suppressing GI cyclins expression and reducing CDKs activity, thus inducing a cell cycle block. We also showed that TAM67 induces apoptosis in serum-free condition in breast cancer cells. DTIC ADA369304: Role of cdc25 Phosphatases in Cellular Immortalization.The goal of this proposal is to study the potential role of tyrosine phosphorylation-dependent checkpoint and especially the cdc25 phosphatase family of cyclin dependent kinase (CDK) activators in cellular immortalization. We have shown that cdc25 phosphatases did in fact extend the normal mammary epithelial cells life span, but do not immortalize them. The effect was most prominent with cdc25A and cdc25B phosphatases. Cdc25C phosphatase' had less effect on HMEC life span. Experiments with catalytically inactive cdc25A mutant show that phosphatase activity of cdc25A is essential for the life span extension. Abstract Background: Infantile hemangioma (IH) is a benign vascular neoplasm that arises from the abnormal proliferation of endothelial cells and enhanced angiogenesis. Results: In the present study, we investigated the β-adrenergic signaling that is associated with hemangioma-derived endothelial cell (HemEC) proliferation. Abstract Background: The major cell cycle control acting at the G2 to mitosis transition is triggered in all eukaryotes by cyclin-dependent kinases (CDKs). In the fission yeast Schizosaccharomyces pombe the activation of the G2/M CDK is regulated primarily by dephosphorylation of the conserved residue Tyr15 in response to the stress-nutritional response and cell geometry sensing pathways. To obtain a more complete view of the G2/M control we have screened systematically for gene deletions that advance cells prematurely into mitosis. Results: A screen of 82% of fission yeast non-essential genes, comprising approximately 3,000 gene deletion mutants, identified 18 genes that act negatively at mitotic entry, 7 of which have not been previously described as cell cycle regulators.
