We have optimized protocols for labeling proteins with P-32 for two/two-dimensional gel electrophoresis. So far, we have identified and characterized one important in vivo substrate, Sic 1. Other workers have shown that an analogous substrate, p27, is important in human cells. The retinoblastoma protein (pRb) is one negative regulator of cellular proliferation. Many breast cancers retain functional pRb and therefore must use other mechanisms to alleviate the tumor-suppressive function of pRb. One common mechanism to circumvent pRb function is amplification of the G1 phase cycling dependent kinases (cdk), cdk4 and cdk6. Cdk4 and cdk6 are thought to have redundant functions as pRb regulators, despite an increasing body of evidence suggesting that certain tumor types specifically activate either cdk4 or cdk6. Regulators of G1 progression including cyclin D1 and the cyclin-dependent kinase inhibitors p21 and p27 appear to be of particular importance in the pathogenesis of breast cancer. We suggest that aggressive breast cancer will involve both (i) overexpression of cyclin D1 and (ii) the failure to undergo a compensatory upregulation of cdk inhibitors. | DTIC ADA470987: The Role of Sphingosine Kinase 2 in Apoptosis of Human Breast Cancer Cells | Two isoforms of sphingosine kinase (SphK) catalyze the formation of sphingosine-1-phosphate (S1P). Whereas, SphK1 stimulates cell growth and survival, it was found that when overexpressed in mouse NIH 3T3 fibroblasts SphK2 enhances caspase-dependent apoptosis in response to serum deprivation, independently of S1P receptors. Sequence analysis revealed that SphK2 contains a 9 amino acid motif similar to that present in BH3-only proteins. Studies showed that the BH3-only domain and catalytic activity contribute to the apoptotic effects of overexpressed SphK2.
