DTIC ADA360338: Development and Use of Novel Tools to Directly Screen for Substrates of Cyclin Dependent KinasesCell division is a central and primary biological function. Many problems in biology and medicine center around cell division, but without a knowledge of the mechanism by which division is controlled, these problems are difficult to attack. The major transitions of the cell cycle are is controlled by a protein called a cyclin dependent kinase, or Cdk. The cell cycles of both yeast and human cells are controlled by a Cdk. Using the yeast two hybrid system I have isolated many novel Cdk interacting proteins in yeast, and these may be instrumental in controlling cell cycle progression. I have also isolate two exciting proteins which can only interact with a substrate trapping mutant of a yeast Cdk. These may be Cdk new substrates. I have also used the genomic technology of Gene Chips, to define the complete set of cell cycle regulated genes in yeast, and to characterize their controlling promoter elements. The target p16 is a tumor suppressor and a CDK inhibitor and is inactivated by gene methylation. The drug 5-aza-2'deoxycytidine (5-Aza-CdR), an inhibitor of DNA methylation, is targeted for reexpression of a repressed p16 in late stage breast cancer patients. Re-activation of the p16 gene in cells in which the gene is methylated will restore normal growth control and be efficacious in treatment of breast cancer. DTIC ADA369304: Role of cdc25 Phosphatases in Cellular Immortalization.The goal of this proposal is to study the potential role of tyrosine phosphorylation-dependent checkpoint and especially the cdc25 phosphatase family of cyclin dependent kinase (CDK) activators in cellular immortalization. We have shown that cdc25 phosphatases did in fact extend the normal mammary epithelial cells life span, but do not immortalize them. The effect was most prominent with cdc25A and cdc25B phosphatases. Cdc25C phosphatase' had less effect on HMEC life span. Experiments with catalytically inactive cdc25A mutant show that phosphatase activity of cdc25A is essential for the life span extension. Our experiments shows that telomerase is not activated by cdc25A expression in normal human cells, suggesting that cdc25 most likely affect Ml. but not M2 checkpoints.19992018-04-25T22:23:31Z

Abstract The development of new small molecule-based therapeutic drugs requires accurate quantification of drug bioavailability, biological activity and treatment efficacy. Rapidly measuring these endpoints is often hampered by the lack of efficient assay platforms with high sensitivity and specificity. Using an in vivo model system, we report a simple and sensitive liquid chromatography-tandem mass spectrometry assay to quantify the bioavailability of a recently developed novel cyclin-dependent kinase inhibitor VMY-1-103, a purvalanol B-based analog whose biological activity is enhanced via dansylation. This study aimed to characterize the CDK4 immunoreactivity in different histological grades of oral leukoplakias (OLs) and oral squamous cell carcinomas (OSCCs) and also aims to discuss its probable role in the tumor biogenesis. Materials and Methods:: Expression of CDK4 was investigated in total of 52 samples including OL (15), OSCCs (30) and normal oral tissues (07). A labeled Streptavidin-Biotin immunohistochemistry assay was performed and staining intensity was evaluated. Results:: The staining pattern was similar in all tissues and was located in both nuclei and cytoplasm. ICP22 is known to reduce the global level of serine (Ser)2 phosphorylation of the Tyr1Ser2Pro3Thr4Ser5Pro6Ser7 heptapeptide repeats comprising the carboxy-terminal domain (CTD) of the large subunit of RNA polymerase (pol) II. Accordingly, ICP22 is thought to associate with and inhibit the activity of the positive-transcription elongation factor b (P-TEFb) pol II CTD Ser2 kinase. We show here that ICP22 causes loss of CTD Ser2 phosphorylation from pol II engaged in transcription of protein-coding genes following ectopic expression in HeLa cells and that recombinant ICP22 interacts with the CDK9 subunit of recombinant P-TEFb. DTIC ADA474451: Mechanism of Antineoplastic Action of Bisperoxovanadium CompoundThis work describes studies into the mechanism of action of bisperoxovanadium (bpVs) compounds and into the basic biology and regulation of Cdc25A phosphatase. The bpVs cause phase-specific cell cycle arrest (G1/S), dose-dependent inhibition of Cdk activity, and persistent Rb hypophosphorylation upon release from serum starvation, consistent with Cdc25A inhibition. Further, they cause p53-independent apoptosis. Oxidative stress and DNA damage do not appear to be involved in their mechanism of action, given that: p53 is not induced by bpV(Me2Phen); p53 and p21 status do not affect IC50; depletion of glutathione or supplementation with antioxidants does not affect IC50, in contrast to other heavy metal-based agents. With respect to regulation, the Cdc25A nuclear localization signal (NLS) was identified and characterized. Suberoylanilide hydroxamic acid, an inhibitor of histone deacetylase, suppresses vasculogenic mimicry and proliferation of highly aggressive pancreatic cancer PaTu8988 cells.This article is from BMC Cancer , volume 14 . Abstract Background: Pancreatic cancer is one of the most aggressive human malignancies with a extremely low 5-year survival rate. Hence, the search for more effective anti-pancreatic cancer agents is urgent. Methods: PaTu8988 pancreatic cancer cells were treated with different concentrations of suberoylanilide hydroxamic acid (SAHA), cell survival, proliferation, migration and vasculogenic mimicry (VM) were analyzed. Associated signaling changes were also analyzed by RT-PCR and Western blots. Results: Here, we reported that SAHA, a histone deacetylase inhibitor (HDACi), exerted significant inhibitory efficiency against pancreatic cancer cell survival, proliferation, migration and VM. SAHA dose-dependently inhibited PaTu8988 pancreatic cancer cell growth with the IC-50 of 3.4 ± 0. 7 μM. DTIC ADA435854: The Role of Ubiquitin E3 Ligase SCFSKP2 in Prostate Cancer DevelopmentThe CDK inhibitor p27Kipl acts as a negative regulator in the cell cycle and in prostate tumorigenesis. Either the loss of p27 or over-expression of its key regulator ubiquitin E3 ligase SKP2 is associated with prostate cancer. Prostate-sepcific expression of SKP2 is sufficient to induce hyperplasia, displasia, and low grade carcinoma in the mouse prostate gland. We propose to examine whether SKP2 cooperates with or is regulated by tumor suppressors such as Pten or Nkx3.1 implicated in prostatic tumorigenesis. In the past year, we have initiated the experiments by breeding and expansion of the colonies, crossing the SKP2 transgenic mice into Pten +/- strains, and ordering Nkx3.1 mice. Compound mice of SKP2 transgene and Pten +/- were made. Our initial studies suggest that there i&. no substantial difference in tumor frequency and grade in the SKP2 and Pten +/- compound mice versus single genetic alterations. The role of ?-adrenergic receptor signaling in the proliferation of hemangioma-derived endothelial cells.This article is from Cell Division , volume 8 . Abstract Background: Infantile hemangioma (IH) is a benign vascular neoplasm that arises from the abnormal proliferation of endothelial cells and enhanced angiogenesis. Recently, propranolol has been found to be effective in the management of IH, suggesting that β-adrenergic receptors (β-ARs) may play an important role in the pathogenesis of IH. Results: In the present study, we investigated the β-adrenergic signaling that is associated with hemangioma-derived endothelial cell (HemEC) proliferation. The results showed that both β1- and β2-ARs were expressed in HemECs. Stimulation of the β-ARs by isoprenaline induced cell proliferation and elevation of second messenger cAMP levels. The proliferation-promoting action of isoprenaline was abolished by a β1-selective antagonist and was more effectively abolished by a β2-selective antagonist; the mechanism for the action of the antagonists was a G0/G1 phase cell cycle arrest which was a;20132014-10-29T13:12:50Z
