| High p27 protein levels in chronic lymphocytic leukemia are associated to low Myc and Skp2 expression, confer resistance to apoptosis and antagonize Myc effects on cell cycle. | This article is from Oncotarget , volume 5 . Abstract Myc (c-Myc) counteracts p27 effects, and low p27 usually correlates with high Myc expression in human cancer. However there is no information on the co-expression of both genes in chronic lymphocytic leukemia (CLL). We found a lack of correlation between RNA and protein levels of p27 and Myc in CLL cells, so we determined the protein levels by immunoblot in 107 cases of CLL. We observed a high p27 protein expression in CLL compared to normal B cells. Ectopic p27 expression in a CLL-derived cell line resulted in cell death resistance. Surprisingly, Myc expression was very low or undetectable in most CLL cases analyzed, with a clear correlation between high p27 and low Myc protein levels. Abstract NUAK1 (NUAK family SnF1-like kinase-1) and NUAK2 protein kinases are activated by the LKB1 tumour suppressor and have been implicated in regulating multiple processes such as cell survival, senescence, adhesion and polarity. In the present paper we present evidence that expression of NUAK1 is controlled by CDK (cyclin-dependent kinase), PLK (Polo kinase) and the SCFβTrCP (Skp, Cullin and F-boxβTrCP) E3 ubiquitin ligase complex. Our data indicate that CDK phosphorylates NUAK1 at Ser445, triggering binding to PLK, which subsequently phosphorylates NUAK1 at two conserved non-catalytic serine residues (Ser476 and Ser480). We have shown that cdc25 phosphatases did in fact extend the normal mammary epithelial cells (HMEC) life span, but do not immortalize them. | DTIC ADA416606: Mapping Critical DNA Sequence Elements Required for Amplification of erbB2 in Breast Cancer | To study the implications of replication initiator proteins to gene amplification, we characterized two proteins required for DNA replication (ORC and Mcm10). In an attempt to purify recombinant human ORC from insect cells infected with baculoviruses expressing HsORC subunits, we found ORC2, -3, -4, and -5 forms a core complex. Expression of ORC3N, which inhibits ORC2-ORC3 interaction, causes cell cycle arrest. Consistent with this result, ORC2-depletion by RNAi arrested cell cycle in G1 with low CDK activity. We propose that cells have a novel mechanism to ensure that CDK2 is activated only after enough pre-RC is formed. We also report that chromatin-loading of Mcm10 requires a functional pre-RC but is independent of Cdc7 activity. Mcm10 is required for chromatin-binding of Cdc45 and origin unwinding. Genistein has been shown to exert chemopreventive and antiproliferative in animal models of cancer and breast cancer cells respectively. Accumulating evidence suggest that genistein is oxidatively metabolized by the cytochrome P45O (CYP45O) family of enzymes, and that its metabolism is specific to breast cancer cells but not nontransformed breast epithelial cells. We investigated the cellular association and effects of metabolism of genistein by CYP45O in T47D breast cancer cells and MCFlOA breast epithelial cells.
