DTIC ADA412793: Chemotherapy of Late Stage Breast Cancer Targeted Towards Cell Cycle Regulatory ComponentsOur original goal was to target cell cycle regulatory components as a potential therapy for breast cancer. The target p16 is a tumor suppressor and a CDK inhibitor and is inactivated by gene methylation. The drug 5-aza-2'deoxycytidine (5-Aza-CdR), an inhibitor of DNA methylation, is targeted for reexpression of a repressed p16 in late stage breast cancer patients. Re-activation of the p16 gene in cells in which the gene is methylated will restore normal growth control and be efficacious in treatment of breast cancer. DTIC ADA360338: Development and Use of Novel Tools to Directly Screen for Substrates of Cyclin Dependent KinasesCell division is a central and primary biological function. Many problems in biology and medicine center around cell division, but without a knowledge of the mechanism by which division is controlled, these problems are difficult to attack. The major transitions of the cell cycle are is controlled by a protein called a cyclin dependent kinase, or Cdk. The cell cycles of both yeast and human cells are controlled by a Cdk. Using the yeast two hybrid system I have isolated many novel Cdk interacting proteins in yeast, and these may be instrumental in controlling cell cycle progression. I have also isolate two exciting proteins which can only interact with a substrate trapping mutant of a yeast Cdk. These may be Cdk new substrates. I have also used the genomic technology of Gene Chips, to define the complete set of cell cycle regulated genes in yeast, and to characterize their controlling promoter elements. DTIC ADA411262: Targeting the AP-1 Transcription Factor for the Treatment of Breast CancerThe AP-l transcription factor is a central component of many signal transduction pathways. We have shown that blocking AP-l by over-expressing a dominant negative form - of cJun (cjun-DN, Tam67) inhibits breast cancer cell growth. We hypothesize that- inhibition of AP-l blocks the cell cycle, reverses TAM-resistance of breast cancer cells, and suppression of AP-l in vivo causes regression of existing breast tumors. In the present study, we demonstrated that TAM67 inhibits breast cancer growth both in vitro and in vivo. We determined the mechanism by which AP-l blockade inhibits breast cancer growth. Our studies suggested that TAM67 inhibits breast cancer growth predominantly by inducing CDK inhibitors (such as P27), suppressing GI cyclins expression and reducing CDKs activity, thus inducing a cell cycle block. We also showed that TAM67 induces apoptosis in serum-free condition in breast cancer cells. Elucidating RUNX protein interactions will help extend our understanding of their roles in normal development and tumorigenesis. In this study, we performed large-scale RNAi screening to identify genes that genetically interact with rnt-1, the sole homolog of RUNX protein in the nematode Caenorhabditis elegans. To this end, we took advantage of the fact that C. elegans can survive a severe loss of RNT-1 function with only mild phenotypes, and we looked for genes that caused a synthetic phenotype in the rnt-1 mutant background. Abstract Objective: Vascular smooth muscle cell (VSMC) hypertrophy and proliferation occur in response to strain-induced local and systemic inflammatory cytokines and growth factors which may contribute to hypertension, atherosclerosis, and restenosis. We hypothesize VSMC strain, modeling normotensive arterial pressure waveforms in vitro, results in attenuated proliferative and increased hypertrophic responses 48 hrs post-strain. Methods: Using Flexcell Bioflex Systems we determined the morphological, hyperplastic and hypertrophic responses of non-strained and biomechanically strained cultured rat A7R5 VSMC. We measured secretion of nitric oxide, key cytokine/growth factors and intracellular mediators involved in VSMC proliferation via fluorescence spectroscopy and protein microarrays. DTIC ADA415783: Regulated Transformation of Mammary Epithelial Cells by Transforming Growth Factor Beta 1We report that MMTV-DNIIR female mice have accelerated mammary gland differentiation during early pregnancy with impaired development during late pregnancy and lactation followed by delayed post-lactational involution. Mammary tumors, mostly carcinoma in situ, developed spontaneously in the MMTV-DNIIR mice with a long median latency (27.5 months). Invading carcinoma cells in MMTV-DNIIR animals showed loss of DNIIR transgene expression as determined by in situ hybridization. The data indicate that signaling from endogenous TGF-Betas not only plays an important role in normal mammary gland physiology but also can also suppress the early stage of tumor formation and contribute to tumor invasion once carcinomas have developed. The ability of the TGF-beta signaling pathways to inhibit proliferation of many epithelial cells while stimulating proliferation fibroblasts remains a conundrum. Abstract Background: Infantile hemangioma (IH) is a benign vascular neoplasm that arises from the abnormal proliferation of endothelial cells and enhanced angiogenesis. Recently, propranolol has been found to be effective in the management of IH, suggesting that β-adrenergic receptors (β-ARs) may play an important role in the pathogenesis of IH. Results: In the present study, we investigated the β-adrenergic signaling that is associated with hemangioma-derived endothelial cell (HemEC) proliferation. The results showed that both β1- and β2-ARs were expressed in HemECs. Stimulation of the β-ARs by isoprenaline induced cell proliferation and elevation of second messenger cAMP levels. DTIC ADA388443: Development and Use of Novel Tools to Directly Screen for Substrates of Cyclin Dependent KinasesThe aim of this research is to gain further insights into the means by which cell proliferation is controlled by cyclin dependent kinases. To this end, a screen has been carried out for proteins that interact with the budding yeast cdk, Cdc28. 10 novel interacting proteins have been identified in a two-hybrid screen using a Gal4/Cdc28 fusion as the bait. Several of the putative interacting proteins have been shown to interact with specific cyclin-Cdc28 kinase complexes. For each of the 10 two-hybrid positives the nature of the Cdc28 interaction is currently being determined by biochemical means. The functional difference between cyclins in their abilities to induce and repress transcription has been investigated using the genomic technology of microarrays. In addition, the complete set of cell cycle regulated genes in yeast has been defined. A systematic screen reveals new elements acting at the G2/M cell cycle control.This article is from Genome Biology , volume 13 . Abstract Background: The major cell cycle control acting at the G2 to mitosis transition is triggered in all eukaryotes by cyclin-dependent kinases (CDKs). In the fission yeast Schizosaccharomyces pombe the activation of the G2/M CDK is regulated primarily by dephosphorylation of the conserved residue Tyr15 in response to the stress-nutritional response and cell geometry sensing pathways. To obtain a more complete view of the G2/M control we have screened systematically for gene deletions that advance cells prematurely into mitosis. Results: A screen of 82% of fission yeast non-essential genes, comprising approximately 3,000 gene deletion mutants, identified 18 genes that act negatively at mitotic entry, 7 of which have not been previously described as cell cycle regulators.
