| DTIC ADA384004: Regulation of Ubiquitin Mediated Proteolysis of G1 Cyclins and the CDK Inhibitor p27 by the Cullin Gene Family in Normal and Tumorigenic Human Breast Cells | Sequential activation and inactivation of cyclin dependent kinases (CDK) regulate eukaryotic cell cycle transitions. The periodicity of CDK activity provides a molecular basis for unidirectional cell cycle progression and is partly controlled by ubiquitin-mediated proteolysis of both cyclins and CDK inhibitors. Deregulated expression of G1 CDK inhibitors and G1 cyclins have been directly linked to breast cancer development. The mechanisms regulating the ubiquitination of these two protein families constitute essential components of G1 cell cycle control in mammalian cells, but they are poorly understood at present. Polyubiquitination of proteins is known to be catalyzed by a cascade of enzymes: El (ubiquitin activating), E2 (ubiquitin conjugating), and E3 (ubiquitin ligating). Two issues critical to our understanding of the regulation of protein turnover are how E3 ligases target proteins for ubiquitination and how they themselves are regulated. | DTIC ADA374391: Oncogenic Functions of cdK4 and cdK6 | Cancer cells often contain disruptions in the transition from G1 phase to S phase. The retinoblastoma protein (pRb) is one negative regulator of cellular proliferation. Many breast cancers retain functional pRb and therefore must use other mechanisms to alleviate the tumor-suppressive function of pRb. One common mechanism to circumvent pRb function is amplification of the G1 phase cycling dependent kinases (cdk), cdk4 and cdk6. Cdk4 and cdk6 are thought to have redundant functions as pRb regulators, despite an increasing body of evidence suggesting that certain tumor types specifically activate either cdk4 or cdk6. This implies that either the function or the regulation of these kinases may be different in different cell types. In an attempt to purify recombinant human ORC from insect cells infected with baculoviruses expressing HsORC subunits, we found ORC2, -3, -4, and -5 forms a core complex. Expression of ORC3N, which inhibits ORC2-ORC3 interaction, causes cell cycle arrest. Consistent with this result, ORC2-depletion by RNAi arrested cell cycle in G1 with low CDK activity. We propose that cells have a novel mechanism to ensure that CDK2 is activated only after enough pre-RC is formed.
