Re-thinking cell cycle regulators: the cross-talk with metabolism.This article is from Frontiers in Oncology , volume 3 . Abstract Analysis of genetically engineered mice deficient in cell cycle regulators, including E2F1, cdk4, and pRB, showed that the major phenotypes are metabolic perturbations. These key cell cycle regulators contribute to lipid synthesis, glucose production, insulin secretion, and glycolytic metabolism. It has been shown that deregulation of these pathways can lead to metabolic perturbations and related metabolic diseases, such as obesity and type II diabetes. The cyclin–cdk–Rb–E2F1 pathway regulates adipogenesis in addition to its well-described roles in cell cycle regulation and cancer. It was also shown that E2F1 directly participates in the regulation of pancreatic growth and function. Similarly, cyclin D3, cdk4, and cdk9 are also adipogenic factors with strong effects on whole organism metabolism. Molecular mechanisms of anti-tumor properties of P276-00 in head and neck squamous cell carcinoma.This article is from Journal of Translational Medicine , volume 11 . Abstract Background: Tumors of the head and neck present aggressive pathological behavior in patients due to high expression of CDK/CCND1 proteins. P276-00, a novel CDK inhibitor currently being tested in clinic, inhibits growth of several cancers in vitro and in vivo. The pre clinical activity of P276-00 in head and neck cancer and its potential mechanisms of action at molecular level are the focus of the current studies. Method: We have investigated the anti-cancer activity of P276-00 in head and neck tumors in vitro and in vivo. DTIC ADA316821: Cellular Proteins Interacting with the Tumor Suppressor Protein p53.Tumor suppressor protein p53 interacts directly with the DNA replication factor RPA and inhibits its ability to bind single-strand-DNA. We defined the domain of p53 that bound to RPA and constructed p53 mutants that failed to bind RPA, but still functioned as transcriptional activators. We found that while these mutants of p53 lost their ability to bind RPA; they still maintained the growth suppression function of p53. Growth suppression function of p53 is dependent on its transactivation activity, probably by inducing p21 and other cell cycle inhibitors. We have extended our study to the p2l protein, which is induced by p53 and interacts with both the cdk2 Icinase and a DNA replication factor PcNA. Here we have demonstrated the importance of PCNA-inhibitory domain of p21 in vivo. DTIC ADA359281: Role of the Cdk Inhibitor Sic 1 in StartMpst cells have four fundamental goals; they must replicate their DNA, segregate their chromosomes, double their mass, and divide. Coordination of these events is essential. I have used the budding yeast S. cerevisiae to dissect the molecular events that control cell cycle progression. I have shown that not only are G1 cyclin-CDK complexes highly unstable rate-limiting activators for cell cycle progression, but that their essential function is to regulate the initiation of DNA replication by phosphorylating and promoting the degradation of a key cell cycle inhibitor, Sic1 (1,2). In fact, cyclin-CDK-regulated proteolysis may constitute the major mechanism of cell cycle control (3). Moreover, Sic1 appears to be the analog of the RB tumor suppressor gene. DTIC ADA435854: The Role of Ubiquitin E3 Ligase SCFSKP2 in Prostate Cancer DevelopmentThe CDK inhibitor p27Kipl acts as a negative regulator in the cell cycle and in prostate tumorigenesis. Either the loss of p27 or over-expression of its key regulator ubiquitin E3 ligase SKP2 is associated with prostate cancer. Prostate-sepcific expression of SKP2 is sufficient to induce hyperplasia, displasia, and low grade carcinoma in the mouse prostate gland. We propose to examine whether SKP2 cooperates with or is regulated by tumor suppressors such as Pten or Nkx3.1 implicated in prostatic tumorigenesis. In the past year, we have initiated the experiments by breeding and expansion of the colonies, crossing the SKP2 transgenic mice into Pten +/- strains, and ordering Nkx3.1 mice. Compound mice of SKP2 transgene and Pten +/- were made. Our initial studies suggest that there i&. no substantial difference in tumor frequency and grade in the SKP2 and Pten +/- compound mice versus single genetic alterations. In addition, in breast cancers where BRCA1 is not mutated, it is often functionally inactivated. Furthermore, cyclin D1 has been shown to be overexpressed in many cancers including breast cancer and its associates with BRCA1. Because of the crucial role of both of these proteins in cancer, it is reasonable to expect that this interaction has a significant role in tumor cells. The understanding of when this interaction occurs during cell cycle progression will help to determine the role of cyclin D1/BRCA1 binding in breast cancer cells. DTIC ADA378108: Role of the Cdk Inhibitor Sic 1 in StartNearly 180,000 women will develop breast cancer in 1999 and 44,000 will die. Cancer is essentially a disease of inappropriate proliferation due to a loss of normal cell cycle controls. We study cell cycle controls in the budding yeast S. cerevisiae. We have shown that the formation of active G1-cyclin dependent kinase complexes (Cdks) is rate limiting for cell cycle progression. The activity of these complexes is regulated through their association with highly unstable G1 cyclins. G1 cyclins are constitutively unstable, and their only essential function is to phosphorylate the B-type Cdk inhibitor, Sic1, and target it for degradation. Sic1 is one of few known in vivo substrates of yeast G1-Cdks. The amount and phosphorylation state of Sic1 appears to determine the timing and size of cell division. Sic1 mutants lacking several phosphorylation sites are stabilized and completely block cell cycle progression. The bpVs cause phase-specific cell cycle arrest (G1/S), dose-dependent inhibition of Cdk activity, and persistent Rb hypophosphorylation upon release from serum starvation, consistent with Cdc25A inhibition. Further, they cause p53-independent apoptosis. Mammary tumors, mostly carcinoma in situ, developed spontaneously in the MMTV-DNIIR mice with a long median latency (27.5 months). Invading carcinoma cells in MMTV-DNIIR animals showed loss of DNIIR transgene expression as determined by in situ hybridization. The data indicate that signaling from endogenous TGF-Betas not only plays an important role in normal mammary gland physiology but also can also suppress the early stage of tumor formation and contribute to tumor invasion once carcinomas have developed.
