Many problems in biology and medicine center around cell division, but without a knowledge of the mechanism by which division is controlled, these problems are difficult to attack. The major transitions of the cell cycle are is controlled by a protein called a cyclin dependent kinase, or Cdk. The cell cycles of both yeast and human cells are controlled by a Cdk. Using the yeast two hybrid system I have isolated many novel Cdk interacting proteins in yeast, and these may be instrumental in controlling cell cycle progression. I have also isolate two exciting proteins which can only interact with a substrate trapping mutant of a yeast Cdk. These may be Cdk new substrates. We have shown that blocking AP-l by over-expressing a dominant negative form - of cJun (cjun-DN, Tam67) inhibits breast cancer cell growth. We hypothesize that- inhibition of AP-l blocks the cell cycle, reverses TAM-resistance of breast cancer cells, and suppression of AP-l in vivo causes regression of existing breast tumors. In the present study, we demonstrated that TAM67 inhibits breast cancer growth both in vitro and in vivo. We determined the mechanism by which AP-l blockade inhibits breast cancer growth. The role of ?-adrenergic receptor signaling in the proliferation of hemangioma-derived endothelial cells. | This article is from Cell Division , volume 8 . Abstract Background: Infantile hemangioma (IH) is a benign vascular neoplasm that arises from the abnormal proliferation of endothelial cells and enhanced angiogenesis. Recently, propranolol has been found to be effective in the management of IH, suggesting that β-adrenergic receptors (β-ARs) may play an important role in the pathogenesis of IH. Results: In the present study, we investigated the β-adrenergic signaling that is associated with hemangioma-derived endothelial cell (HemEC) proliferation. The results showed that both β1- and β2-ARs were expressed in HemECs. Stimulation of the β-ARs by isoprenaline induced cell proliferation and elevation of second messenger cAMP levels. DTIC ADA388443: Development and Use of Novel Tools to Directly Screen for Substrates of Cyclin Dependent Kinases | The aim of this research is to gain further insights into the means by which cell proliferation is controlled by cyclin dependent kinases. To this end, a screen has been carried out for proteins that interact with the budding yeast cdk, Cdc28. 10 novel interacting proteins have been identified in a two-hybrid screen using a Gal4/Cdc28 fusion as the bait. Several of the putative interacting proteins have been shown to interact with specific cyclin-Cdc28 kinase complexes. For each of the 10 two-hybrid positives the nature of the Cdc28 interaction is currently being determined by biochemical means. The functional difference between cyclins in their abilities to induce and repress transcription has been investigated using the genomic technology of microarrays. In addition, the complete set of cell cycle regulated genes in yeast has been defined. A systematic screen reveals new elements acting at the G2/M cell cycle control. | This article is from Genome Biology , volume 13 . Abstract Background: The major cell cycle control acting at the G2 to mitosis transition is triggered in all eukaryotes by cyclin-dependent kinases (CDKs). In the fission yeast Schizosaccharomyces pombe the activation of the G2/M CDK is regulated primarily by dephosphorylation of the conserved residue Tyr15 in response to the stress-nutritional response and cell geometry sensing pathways. To obtain a more complete view of the G2/M control we have screened systematically for gene deletions that advance cells prematurely into mitosis. Results: A screen of 82% of fission yeast non-essential genes, comprising approximately 3,000 gene deletion mutants, identified 18 genes that act negatively at mitotic entry, 7 of which have not been previously described as cell cycle regulators.
