To this end, a screen has been carried out for proteins that interact with the budding yeast cdk, Cdc28. 10 novel interacting proteins have been identified in a two-hybrid screen using a Ga14/Cdc28 fusion as the bait. Several of the putative interacting proteins have been shown to interact with specific cyclin-Cdc28 kinase complexes. For each of the 10 two-hybrid positives the nature of the Cdc28 interaction is currently being determined by biochemical means. | DTIC ADA392320: Role of cdc25 Phosphatases in Cellular Immortalization | The goal of this proposal is to study the potential role of tyrosine phosphorylation-dependent checkpoint and especially the cdc25 phosphatase family of cyclin dependent kinase (CDK) activators in cellular immortalization. We have shown that cdc25 phosphatases did in fact extend the normal mammary epithelial cells (HMEC) life span, but did not immortalize them. The effect was most prominent with cdc25A and cdc25B phosphatases. Cdc25C phosphatase had less effect on HMEC life span. Experiments with catalytically inactive cdc25A mutant show that phosphatase activity of cdc2SA is essential for the life span extension. Our experiments show that telomerase is not activated by cdc25A expression in normal human cells, suggesting that cdc25 most likely affect Ml, but not M2 checkpoints. We have explored the signalling pathways and the molecular mechanisms involved. Either activated Ras or an inducible Raf onco-protein alone, induce a Gi-specific cell cyle arrest. This requires prolonged activation of MAP kinase activity followed by the inhibition of cyclin-dependent kinases (cdk) via induction of the cyclin/cdk inhibitory protein, p21 Cipi. Abstract The CDK/Rb/E2F pathway is commonly disrupted in lung cancer, and thus, it is predicted that blocking the E2F pathway would have therapeutic potential. To test this hypothesis, we have examined the activity of HLM006474 (a small molecule pan-E2F inhibitor) in lung cancer cell lines as a single agent and in combination with other compounds. HLM006474 reduces the viability of both SCLC and NSCLC lines with a biological IC50 that varies between 15 and 75 µM, but with no significant difference between the groups. Combination of HLM006474 with cisplatin and gemcitabine demonstrate little synergy; however, HLM006474 synergizes with paclitaxel. Surprisingly, we discovered that brief treatment of cells with HLM006474 led to an increase of E2F3 protein levels (due to de-repression of these promoter sites).
