| MolClass: a web portal to interrogate diverse small molecule screen datasets with different computational models. | This article is from Bioinformatics , volume 28 . Abstract Summary: The MolClass toolkit and data portal generate computational models from user-defined small molecule datasets based on structural features identified in hit and non-hit molecules in different screens. Each new model is applied to all datasets in the database to classify compound specificity. MolClass thus defines a likelihood value for each compound entry and creates an activity fingerprint across diverse sets of screens. MolClass uses a variety of machine-learning methods to find molecular patterns and can therefore also assign a priori predictions of bioactivities for previously untested molecules. We defined the domain of p53 that bound to RPA and constructed p53 mutants that failed to bind RPA, but still functioned as transcriptional activators. We found that while these mutants of p53 lost their ability to bind RPA; they still maintained the growth suppression function of p53. We have shown that the formation of active G1-cyclin dependent kinase complexes (Cdks) is rate limiting for cell cycle progression. | DTIC ADA455296: Consequences of Cyclin D1/BRCA1 Interaction in Breast Cancer Progression | The inheritance of one defective BRCA1 or BRCA2 allele predisposes an individual to developing breast, ovarian and T-cell cancers. In addition, in breast cancers where BRCA1 is not mutated, it is often functionally inactivated. Furthermore, cyclin D1 has been shown to be overexpressed in many cancers including breast cancer and its associates with BRCA1. Because of the crucial role of both of these proteins in cancer, it is reasonable to expect that this interaction has a significant role in tumor cells. The understanding of when this interaction occurs during cell cycle progression will help to determine the role of cyclin D1/BRCA1 binding in breast cancer cells. Therefore, I hypothesize that the direct interaction of cyclin D1 with BRCA1 results in the cell cycle dependent regulation of the activity of BRCA1.
