| Ep 10 Part 1: Kristine Bunch's Nightmare | In part 1 of this episode, you'll hear about one woman's nightmare come true. In 1995, 21-year-old Kristine Bunch was abruptly awoken in the early morning hours to discover that her house was on fire. After escaping the flames, she watched in horror as firefighters were unable to save her 3-year-old son who was still trapped inside. Within hours of being released from the hospital, she discovered that detectives had a theory about the fire - someone had started the fire on purpose, and their main suspect was Kristine. A special thank you to Kristine Bunch for helping me tell her story. Please consider supporting Court Junkie with as little as $3 a month via Patreon.com/CourtJunkie. | Lawyer Says Bin Laden Did Not Orchestrate 9/11 | Al-Qaeda leader never claimed responsibility for attack, Van de Wiel tells Australian Supreme Court Paul Joseph Watson Prison Planet Thursday, February 28th, 2008 Skepticism towards the 9/11 official story reached the Australian Supreme Court this week after a lawyer asserted that the attacks were not orchestrated by Osama bin Laden during the trial of an alleged terror group in Melbourne. "To say that this was all orchestrated by Osama bin Laden is a silly thing to do. He's never claimed responsibility for it," Remy van de Wiel QC told the Australian jury. | DTIC ADA392320: Role of cdc25 Phosphatases in Cellular Immortalization | The goal of this proposal is to study the potential role of tyrosine phosphorylation-dependent checkpoint and especially the cdc25 phosphatase family of cyclin dependent kinase (CDK) activators in cellular immortalization. We have shown that cdc25 phosphatases did in fact extend the normal mammary epithelial cells (HMEC) life span, but did not immortalize them. The effect was most prominent with cdc25A and cdc25B phosphatases. Cdc25C phosphatase had less effect on HMEC life span. Experiments with catalytically inactive cdc25A mutant show that phosphatase activity of cdc2SA is essential for the life span extension. Our experiments show that telomerase is not activated by cdc25A expression in normal human cells, suggesting that cdc25 most likely affect Ml, but not M2 checkpoints. Retroviral cDNA libraries (sense and anti-sense) were introduced into HMEC together with cdc25 to assay for the potential 'enhancement' of cdc25 ability to expand the cellular life span. | DTIC ADA358053: The p16 Pathway In Breast Cancer and Senescence Control. | The p16INK4a protein, hereafter referred to as pl6, is a known negative regulator of cell cycle progression through its cyclin dependent kinase (CDK) inhibiting function (1). pl6 competes with the activating D type cyclins for association with CDK4 or CDK6, thereby preventing phosphorylation of proteins controlling Ol exit such as the retinoblastoma (Rb) protein (2). Inactivation of p16 has been observed in numerous tumor types (3-8), and lack or reduced expression of p16 has also been shown through a variety of technical approaches to occur in at least 50% of the breast cancer samples examined (9, 10). While these findings suggest that p16 may play an important role in breast tumorigenesis, the consequence of such abberrations of p16 are not yet clear. | 1998 | 2018-04-21T04:36:31Z |

| DTIC ADA474916: Checkpoint Kinase-Dependent Regulation of DNA Repair and Genome Instability in Breast Cancer | DDB1, a component of a Cul4A ubiquitin ligase complex, promotes nucleotide excision repair (NER) and regulates DNA replication. We have investigated the role of human DDB1 in maintaining genome stability. DDB1-depleted cells accumulate DNA double strand breaks in widely dispersed regions throughout the genome and have activated ATM and ATR cell cycle checkpoints. Depletion of Cul4A yields similar phenotypes, indicating that an E3-ligase function of DDB1 is important for genome maintenance. In contrast, depletion of DDB2, XPA, or XPC does not cause activation of DNA damage checkpoints, indicating that defects in NER are not involved. One substrate of DDB1-Cul4A that is crucial for preventing genome instability is Cdt1. DDB1-depleted cells exhibit increased levels of Cdt1 protein and re-replication, despite containing other Cdt1 regulatory mechanisms. | DTIC ADA389563: Role of c-Myc in Estrogen Stimulated Cell Proliferation in Normal and Malignant Breast Epithelial Cells | Estrogenic steroids have diverse physiological functions and are critically involved in the pathogenesis of breast cancer. The transcriptional regulator c-Myc has been implicated in estrogen-induced mitogenesis and we are therefore investigating the contribution of c-Myc to various downstream molecular and cellular events after estrogen stimulation. Transcriptional activation of c-myc is an early response to estrogen treatment and we have previously shown that induction of c-Myc quantitatively reproduces the effects of estrogen on cell cycle progression. Furthermore, c-Myc induction mimics other downstream effects of estrogen including activation of cyclin E-Cdk2 by decreased association with the CDK inhibitor p2l. Accumulating evidence suggests that the transforming capacity of c-Myc correlates more closely with transcriptional repression than with activation, raising the question of which function is the more important for c-Myc regulation of cell cycle progression. | DTIC ADA396497: Use p27KIP1 Degradation for Breast Cancer Diagnosis and Therapy | It is well established that p27(KIP1), a CDK inhibitor, serves as an excellent prognostic marker for breast cancers. Loss or low levels of p27 are closely associated with poor prognosis in breast cancer patients. We have identified a ubiquitin E3 ligase complex, SCF(SKP2), that binds and targets p27 for ubiquitin-dependent proteolysis. We found that SKP2, the rate limiting component of SCF(SKP2) complex, is highly induced in many transformed cells. We proposed to determine whether there is an inverse relationship between the levels of p27 and SKP2 in breast cancer cells and tumor samples. In the past year, we have affinity purified antibodies against the components of SCF(SKP2) including anti-SKP2, SKP1, CUL-1, and p27 antibodies. The affinity purified antibodies were used to stain breast tumor samples. We also began to characterize the determinants on phospho-p27 peptide that mediate interaction between p27 and SKP2. | E2F Inhibition Synergizes with Paclitaxel in Lung Cancer Cell Lines. | This article is from PLoS ONE , volume 9 . Abstract The CDK/Rb/E2F pathway is commonly disrupted in lung cancer, and thus, it is predicted that blocking the E2F pathway would have therapeutic potential. To test this hypothesis, we have examined the activity of HLM006474 (a small molecule pan-E2F inhibitor) in lung cancer cell lines as a single agent and in combination with other compounds. HLM006474 reduces the viability of both SCLC and NSCLC lines with a biological IC50 that varies between 15 and 75 µM, but with no significant difference between the groups. Combination of HLM006474 with cisplatin and gemcitabine demonstrate little synergy; however, HLM006474 synergizes with paclitaxel. Surprisingly, we discovered that brief treatment of cells with HLM006474 led to an increase of E2F3 protein levels (due to de-repression of these promoter sites). Since paclitaxel sensitivity has been shown to correlate with E2F3 levels, we hypothesized that HLM006474 synergy with paclitaxel may be me; | 2014 | 2014-10-22T18:06:02Z |
| Capsaicin-Induced Apoptosis of FaDu Human Pharyngeal Squamous Carcinoma Cells. | This article is from Yonsei Medical Journal , volume 53 . Abstract Purpose: To investigate the anti-tumor effect of capsaicin on human pharyngeal squamous carcinoma cells (FaDu). Materials and Methods: The expression of apoptosis/cell cycle-related proteins (or genes) was examined by reverse transcriptase-polymerase chain reaction, western blotting and ELISA methods, while the apoptotic cell population, cell morphology and DNA fragmentation levels were assessed using flow cytometry, fluorescence microscopy and agarose gel electrophoresis. Results: Capsaicin was found to inhibit the growth and proliferation of FaDu cells in a dose- and time-dependent manner. Apoptotic cell death was confirmed by observing increases in nuclear condensation, nuclear DNA fragmentation and sub-G1 DNA content. The observed increase in cytosolic cytochrome c, activation of caspase 3 and PARP (p85) levels following capsaicin treatment indicated that the apoptotic response was mitochondrial pathway-dependent. | Cyclin-dependent kinases 7 and 9 specifically regulate neutrophil transcription and their inhibition drives apoptosis to promote resolution of inflammation. | This article is from Cell Death and Differentiation , volume 19 . Abstract Terminally differentiated neutrophils are short-lived but the key effector cells of the innate immune response, and have a prominent role in the pathogenesis and propagation of many inflammatory diseases. Delayed apoptosis, which is responsible for their extended longevity, is critically dependent on a balance of intracellular survival versus pro-apoptotic proteins. Here, we elucidate the mechanism by which the cyclin-dependent kinase (CDK) inhibitor drugs such as R-roscovitine and DRB (5,6-dichloro-1-beta-𝒟-ribofuranosylbenzimidazole) mediate neutrophil apoptosis. We demonstrate (by a combination of microarray, confocal microscopy, apoptosis assays and western blotting) that the phosphorylation of RNA polymerase II by CDKs 7 and 9 is inhibited by R-roscovitine and that specific effects on neutrophil transcriptional capacity are responsible for neutrophil apoptosis. Finally, we show that specific CDK7 and 9 i; | 2012 | 2014-10-26T21:30:14Z |
| Roscovitine confers tumor suppressive effect on therapy-resistant breast tumor cells. | This article is from Breast Cancer Research : BCR , volume 13 . Abstract Introduction: Current clinical strategies for treating hormonal breast cancer involve the use of anti-estrogens that block estrogen receptor (ER)α functions and aromatase inhibitors that decrease local and systemic estrogen production. Both of these strategies improve outcomes for ERα-positive breast cancer patients, however, development of therapy resistance remains a major clinical problem. Divergent molecular pathways have been described for this resistant phenotype and interestingly, the majority of downstream events in these resistance pathways converge upon the modulation of cell cycle regulatory proteins including aberrant activation of cyclin dependent kinase 2 (CDK2). In this study, we examined whether the CDK inhibitor roscovitine confers a tumor suppressive effect on therapy-resistant breast epithelial cells. Methods: Using various in vitro and in vivo assays, we tested the effect of roscovitine on th; | 2011 | 2014-10-28T11:55:39Z |
| MyoD regulates p57kip2 expression by interacting with a distant cis-element and modifying a higher order chromatin structure. | This article is from Nucleic Acids Research , volume 40 . Abstract The bHLH transcription factor MyoD, the prototypical master regulator of differentiation, directs a complex program of gene expression during skeletal myogenesis. The up-regulation of the cdk inhibitor p57kip2 plays a critical role in coordinating differentiation and growth arrest during muscle development, as well as in other tissues. p57kip2 displays a highly specific expression pattern and is subject to a complex epigenetic control driving the imprinting of the paternal allele. However, the regulatory mechanisms governing its expression during development are still poorly understood. We have identified an unexpected mechanism by which MyoD regulates p57kip2 transcription in differentiating muscle cells. We show that the induction of p57kip2 requires MyoD binding to a long-distance element located within the imprinting control region KvDMR1 and the consequent release of a chromatin loop involving p57kip2 promoter.
