Predictive value of progesterone receptor in advanced-stage breast cancer patients treated with CDK 4/6 inhibitorsABSTRACT Aims: Phase III studies investigating CDK 4/6 inhibitors have failed to identify significant predictive or prognostic markers that aid clinicians in therapeutic decision-making. Given the complex treatment landscape in breast cancer, identifying patient and tumor characteristics that optimize the utilization of CDK 4/6 inhibitors across diverse therapeutic approaches is crucial. In our study, we aimed to evaluate the predictive role of progesterone receptor (PR) expression levels in patients with estrogen receptor (ER)-positive and human epidermal growth factor receptor 2 (HER2)-negative advanced-stage breast cancer treated with CDK 4/6 inhibitors. Methods: This study retrospectively evaluated 244 patients who received a combination of CDK 4/6 inhibitors and endocrine therapy as their first-line treatment. Assessment of network perturbation amplitudes by applying high-throughput data to causal biological networks.This article is from BMC Systems Biology , volume 6 . Abstract Background: High-throughput measurement technologies produce data sets that have the potential to elucidate the biological impact of disease, drug treatment, and environmental agents on humans. The scientific community faces an ongoing challenge in the analysis of these rich data sources to more accurately characterize biological processes that have been perturbed at the mechanistic level. Here, a new approach is built on previous methodologies in which high-throughput data was interpreted using prior biological knowledge of cause and effect relationships. These relationships are structured into network models that describe specific biological processes, such as inflammatory signaling or cell cycle progression. This enables quantitative assessment of network perturbation in response to a given stimulus. Several pharmacological CDK inhibitors are currently in clinical trials as potential cancer therapeutics and some of them also exhibit antiviral effects. Olomoucine II and roscovitine, purine-based inhibitors of CDKs, were described as effective antiviral agents that inhibit replication of a broad range of wild type human viruses. Olomoucine II and roscovitine show high selectivity for CDK7 and CDK9, with important functions in the regulation of RNA polymerase II transcription. RNA polymerase II is necessary for viral transcription and following replication in cells. We analyzed the effect of inhibition of CDKs by olomoucine II on gene expression from viral promoters and compared its effect to widely-used roscovitine. Abstract Chromatin proteins mediate replication, regulate expression, and ensure integrity of the genome. So far, a comprehensive inventory of interphase chromatin has not been determined. This is largely due to its heterogeneous and dynamic composition, which makes conclusive biochemical purification difficult, if not impossible. As a fuzzy organelle, it defies classical organellar proteomics and cannot be described by a single and ultimate list of protein components. Instead, we propose a new approach that provides a quantitative assessment of a protein's probability to function in chromatin. We integrate chromatin composition over a range of different biochemical and biological conditions. This resulted in interphase chromatin probabilities for 7635 human proteins, including 1840 previously uncharacterized proteins. On The Development of Biophysical Models for Space Radiation Risk AssessmentExperimental techniques in molecular biology are being applied to study biological risks from space radiation. The use of molecular assays presents a challenge to biophysical models which in the past have relied on descriptions of energy deposition and phenomenological treatments of repair. We describe a biochemical kinetics model of cell cycle control and DNA damage response proteins in order to model cellular responses to radiation exposures. Using models of cyclin-cdk, pRB, E2F's, p53, and GI inhibitors we show that simulations of cell cycle populations and GI arrest can be described by our biochemical approach. DTIC ADA316821: Cellular Proteins Interacting with the Tumor Suppressor Protein p53.Tumor suppressor protein p53 interacts directly with the DNA replication factor RPA and inhibits its ability to bind single-strand-DNA. We defined the domain of p53 that bound to RPA and constructed p53 mutants that failed to bind RPA, but still functioned as transcriptional activators. We found that while these mutants of p53 lost their ability to bind RPA; they still maintained the growth suppression function of p53. Growth suppression function of p53 is dependent on its transactivation activity, probably by inducing p21 and other cell cycle inhibitors. We have extended our study to the p2l protein, which is induced by p53 and interacts with both the cdk2 Icinase and a DNA replication factor PcNA. Here we have demonstrated the importance of PCNA-inhibitory domain of p21 in vivo. DTIC ADA435854: The Role of Ubiquitin E3 Ligase SCFSKP2 in Prostate Cancer DevelopmentThe CDK inhibitor p27Kipl acts as a negative regulator in the cell cycle and in prostate tumorigenesis. Either the loss of p27 or over-expression of its key regulator ubiquitin E3 ligase SKP2 is associated with prostate cancer. Prostate-sepcific expression of SKP2 is sufficient to induce hyperplasia, displasia, and low grade carcinoma in the mouse prostate gland. We propose to examine whether SKP2 cooperates with or is regulated by tumor suppressors such as Pten or Nkx3.1 implicated in prostatic tumorigenesis. In the past year, we have initiated the experiments by breeding and expansion of the colonies, crossing the SKP2 transgenic mice into Pten +/- strains, and ordering Nkx3.1 mice. Compound mice of SKP2 transgene and Pten +/- were made. Our initial studies suggest that there i&. no substantial difference in tumor frequency and grade in the SKP2 and Pten +/- compound mice versus single genetic alterations. DTIC ADA378108: Role of the Cdk Inhibitor Sic 1 in StartNearly 180,000 women will develop breast cancer in 1999 and 44,000 will die. Cancer is essentially a disease of inappropriate proliferation due to a loss of normal cell cycle controls. We study cell cycle controls in the budding yeast S. cerevisiae. We have shown that the formation of active G1-cyclin dependent kinase complexes (Cdks) is rate limiting for cell cycle progression. The activity of these complexes is regulated through their association with highly unstable G1 cyclins. G1 cyclins are constitutively unstable, and their only essential function is to phosphorylate the B-type Cdk inhibitor, Sic1, and target it for degradation. Sic1 is one of few known in vivo substrates of yeast G1-Cdks. The amount and phosphorylation state of Sic1 appears to determine the timing and size of cell division. Sic1 mutants lacking several phosphorylation sites are stabilized and completely block cell cycle progression. DTIC ADA482640: Suppression of Prostate Tumors by INK4C and PTENWork during this entire funding period thoroughly characterized mutant mice in the cyclin-dependent kinase (CDK) inhibitor p18Ink4c, which causes hyperplastic cell growth in various tissues including the prostate and the lipid phosphatase Pten, which regulates cell growth and is highly associated with the development of human prostate cancer. The first two years of funding were highly productive and resulted in one manuscript publication supporting a functional collaboration between p18Ink4c and Pten in tumor suppression. Functional inactivation or reduction of this pathway is a common event found in many types of human tumors. DTIC ADA382749: Resistance to Tamoxifen: A Consequence of Altered p27Kipl Regulation During Breast Cancer ProgressionThis grant addresses the molecular mechanisms of resistance to the anti-estrogen, Tamoxifen, in breast cancers. Both estrogens and antiestrogens influence the cell cycle during the GI phase. In breast tumors, loss of the cdk inhibitor, p27, is associated with a poor prognosis and with steroid resistance. We postulate that altered p27 degradation in breast cancer may contribute to resistance to antiestrogen therapy. In the last year, we have assayed cell cycle effects of estradiol in the ER+ breast cancer cell line, MCF-7. MCF-7 arrests in G1 upon removal of estradiol or on addition of Tamoxifen. Upon re-addition of estradiol, cells re-enter the cell cycle with onset of S phase by 12 hours and loss of both p21 and p27 proteins. Cyclin D1 associated kinase activity rose transiently in early G1. Activation of cyclin E/cdk2 kinase in mid-late G1 was accompanied by loss of cyclin E-associated p21 and p27.
