Abstract This report highlights the phytochemical analysis, antioxidant potential and anticancer activity against breast carcinoma of 70% methanolic extract of lichen, Parmotrema reticulatum (PRME). Phytochemical analysis of PRME confirms the presence of various phytoconstituents like alkaloids, carbohydrates, flavonoids, glycosides, phenols, saponins, tannins, anthraquinones, and ascorbic acid; among which alkaloids, phenols and flavonoids are found in abundant amount. High performance liquid chromatography (HPLC) analysis of PRME revealed the presence of catechin, purpurin, tannic acid and reserpine. Antioxidant activity was evaluated by nine separate methods. PRME showed excellent hydroxyl and hypochlorous radical scavenging as well as moderate DPPH, superoxide, singlet oxygen, nitric oxide and peroxynitrite scavenging activity. | DTIC ADA443026: Radiation Sensitization in Breast Cancer via Targeting Survivin Expression | We proposed to determine whether overexpression- of surviving results in radio resistance and the possible mechanisms; whether irradiation down regulates p34(CDC2) and its mechanism in vascular endothelium and whether CDK (cyclin dependent kinase) inhibitors sensitize it to radiation injury; the mechanism of surviving- deregulation in breast cancer and whether inhibition of surviving or its regulator, p34(CDC2) abolishes radio resistance. We found that overexpression of surviving leads to radiation resistance and CDK inhibitors result in radiosensitization. We also found that deregulation of surviving in breast cancer is mediated by Stat3 (Signal transducer and activator of transcription). Abstract DNA double-strand breaks (DSBs) can be repaired by one of two major pathways—non-homologous end-joining (NHEJ) and homologous recombination (HR)—depending on whether cells are in G1 or S/G2 phase, respectively. However, the mechanisms of DSB repair during M phase remain largely unclear. In this study, we demonstrate that transient treatment of M-phase cells with the chemotherapeutic topoisomerase inhibitor etoposide induced DSBs that were often associated with anaphase bridge formation and genome instability such as dicentric chromosomes. Although most of the DSBs were carried over into the next G1 phase, some were repaired during M phase. Both NHEJ and HR, in particular NHEJ, promoted anaphase-bridge formation, suggesting that these repair pathways can induce genome instability during M phase.
