| DTIC ADA392324: Isolation of Proteins Interacting With the Cyclin D1-CDK6 Complex From Normal and Tumorigenic Human Breast Cells Using a Novel Yeast Three-Hybrid System | The eukaryotic cell division cycle is primary controlled by a family of protein serine/threonine kinases, the cyclin-dependent-kinases (CDKs). Activation of CDKs is dependent upon the interaction with an activating cyclin subunit. Presumably, these kinases progress the cell cycle by phosphorylation of critical cellular proteins. The detection of these transient kinase-substrate protein-protein interactions using conventional methods presents a major technical challenge, and is primarily responsible for our poor understanding of CDK substrates. The yeast two-hybrid system is a method for detecting protein-protein interactions. A major limitation of this method is the fact that only binary interactions can be detected. Using a modified version of the two-hybrid system, I am able to detect the interaction of ternary protein complexes. For both cyclin A/cdk2 and cyclin E/cdk2, the presence of a Cy motif decreased the Km(sup peptide! 75 to 120-fold while the kcat remained unchanged. Changes in the length of the linker between the Cy motif and the phosphoacceptor serine suggest that both sites are engaged simultaneously to the cyclin and the cdk, respectively. PS100, a peptide containing a Cy motif, acts as a competitive inhibitor of cyclin/cdk complexes with a 15-fold lower Ki for cyclin E/cdk2 than for cyclin A/cdk2. These results provide kinetic proof that a Cy motif located at a minimal distance from the SPXK is essential for optimal phosphorylation by cdks and suggest that small chemicals that mimic the Cy motif would be specific inhibitors of substrate recognition by cyclin-dependent kinases. We have identified a novel therapeutic strategy for MYCN-amplified neuroblastoma using a novel first-in-class inhibitor of cyclin dependent kinase 7 and have demonstrated selective potent activity against these tumors without general toxicity. Additionally, we have determined that in tumors expressing mutated ALK but without MYCN amplification, the combination of an ALK inhibitor and a transcriptional CDK inhibitor is synergistic. | Exome sequencing reveals a potential mutational trajectory and treatments for a specific pancreatic cancer patient. | This article is from OncoTargets and therapy , volume 7 . Abstract Pancreatic cancer is the fourth biggest killer, and has one of the worst prognoses, of any cancer type. Approximately 95% of patients diagnosed with pancreatic cancer will not survive beyond 5 years post diagnosis, and these statistics have barely improved in over 40 years. Here, genomic changes in one particular patient with stage IV metastatic pancreatic cancer were explored to suggest a potential personalized treatment. In particular, exome sequencing of genomic DNA extracted from blood and the cancer biopsy was utilized with the aim of identifying mutational drivers of the cancer. This analysis revealed a splice site mutation in RBCK1 as the most promising driver of the cancer and a therapy based on a pan-cyclin-dependent kinase (pan-CDK) inhibitor, flavopiridol. However, the roles and molecular mechanisms of CDK4 triggering nasopharynx carcinogenesis are still unclear. Methods: Lentiviral-vector mediated shRNA was used to suppress CDK4 expression and examine its molecular mechanisms. Using immunohistochemistry, we analyzed CDK4 protein expression in clinicopathologically characterized nasopharyngeal carcinoma (NPC) cases and nasopharyngeal tissues (NPs). Survival curves were plotted by the Kaplan-Meier method and compared using the log-rank test. Results: In this investigation, we knocked down CDK4 expression and observed that NPC cell growth and cell cycle progression were significantly blocked by suppressing expression of CCND1, CDK6, and E2F1 as well as elevated p21 expression.
