| Inhibition of Post-Transcriptional RNA Processing by CDK Inhibitors and Its Implication in Anti-Viral Therapy. | This article is from PLoS ONE , volume 9 . Abstract Cyclin-dependent kinases (CDKs) are key regulators of the cell cycle and RNA polymerase II mediated transcription. Several pharmacological CDK inhibitors are currently in clinical trials as potential cancer therapeutics and some of them also exhibit antiviral effects. Olomoucine II and roscovitine, purine-based inhibitors of CDKs, were described as effective antiviral agents that inhibit replication of a broad range of wild type human viruses. Olomoucine II and roscovitine show high selectivity for CDK7 and CDK9, with important functions in the regulation of RNA polymerase II transcription. RNA polymerase II is necessary for viral transcription and following replication in cells. An HA-epitope tagged Rsu-1 cDNA was introduced into the MCF7 breast carcinoma cell line. The transcriptional regulator c-Myc has been implicated in estrogen-induced mitogenesis and we are therefore investigating the contribution of c-Myc to various downstream molecular and cellular events after estrogen stimulation. Transcriptional activation of c-myc is an early response to estrogen treatment and we have previously shown that induction of c-Myc quantitatively reproduces the effects of estrogen on cell cycle progression. Furthermore, c-Myc induction mimics other downstream effects of estrogen including activation of cyclin E-Cdk2 by decreased association with the CDK inhibitor p2l. | Novel structural features of CDK inhibition revealed by an ab initio computational method combined with dynamic simulations | The rational development of specific inhibitors for the ~500 protein kinases encoded in the human genome is impeded by a poor understanding of the structural basis for the activity and selectivity of small molecules that compete for ATP binding. Combining classical dynamic simulations with a novel ab initio computational approach linear-scalable to molecular interactions involving thousands of atoms, we have investigated the binding of five distinct inhibitors to the cyclin-dependent kinase CDK2. We report here that polarization and dynamic hydrogen bonding effects, so far undetected by crystallography, affect both their activity and selectivity. | DTIC ADA357346: Biology of Somatostatin and Somatostatin Receptors in Breast Cancer. | Fifty additional primary human breast tumor samples were analysed for SSTR1-5 mRNA and shown to be all receptor positive. Mean SSTR mRNA level was the highest for SSTR3 followed by SSTR1, SSTR2, SSTR5, and SSTR4. Statistical analysis showed a negative correlation between SSTR2 and tumor size, a positive correlation between SSTR2 and estrogen receptors (ER), and a trend towards a positive correlation between SSTR2 and progesterone receptor, SSTR1 and ER, SSTR1 and lymph node status, and SSTR3 and tumor grade. Breast cancer cell lines also expressed multiple SSTR subtypes but the level of receptor expression was less than that in solid tumors.
