Aberrations in the ubiquitin-mediated destruction pathways, and particularly in the destruction of the Dl cyclin, have been implicated in human breast cancer. A cascade of enzymes leads to ubiquitination of substrate: El enzyme (ubiquitin activating); E2 enzyme (ubiquitin conjugating), and E3 enzyme (ubiquitin ligase). Functional inactivation or reduction of this pathway is a common event found in many types of human tumors. To test how the Rb pathway interacts with other cellular pathways in tumor suppression, we characterized mice with combined mutations in the CDK inhibitor p18 (exp Ink4c) and the lipid phosphatase PTEN, which regulates cell growth and survival. Specifically, we explored the multifactorial nature of the interaction a growth factor (TGFalpha, which is commonly expressed both in benign and malignant proliferative disease of the human breast) and an oncogene (c-myc, whose gene is amplified and whose protein is inappropriately expressed in 20-30% of human breast tumors. Using a transgenic mouse model we observed that co-expression of these two genes was remarkably synergistic for onset and progression of cell survival and proliferation. While mye induced both p53 and bax death-promoting genes, TCFalpha promoted cell survival by inducing Bc1XL. In terms of the cell cycle, mye shortened G1 by modulating cyc1lin E, p27, and cdc25A, which activated cdk-2 and inactivated Rb. | DTIC ADA389563: Role of c-Myc in Estrogen Stimulated Cell Proliferation in Normal and Malignant Breast Epithelial Cells | Estrogenic steroids have diverse physiological functions and are critically involved in the pathogenesis of breast cancer. The transcriptional regulator c-Myc has been implicated in estrogen-induced mitogenesis and we are therefore investigating the contribution of c-Myc to various downstream molecular and cellular events after estrogen stimulation. Transcriptional activation of c-myc is an early response to estrogen treatment and we have previously shown that induction of c-Myc quantitatively reproduces the effects of estrogen on cell cycle progression. Furthermore, c-Myc induction mimics other downstream effects of estrogen including activation of cyclin E-Cdk2 by decreased association with the CDK inhibitor p2l. Accumulating evidence suggests that the transforming capacity of c-Myc correlates more closely with transcriptional repression than with activation, raising the question of which function is the more important for c-Myc regulation of cell cycle progression.
