Timely Activation of Budding Yeast APCCdh1 Involves Degradation of Its Inhibitor, Acm1, by an Unconventional Proteolytic Mechanism.This article is from PLoS ONE , volume 9 . Abstract Regulated proteolysis mediated by the ubiquitin proteasome system is a fundamental and essential feature of the eukaryotic cell division cycle. Most proteins with cell cycle-regulated stability are targeted for degradation by one of two related ubiquitin ligases, the Skp1-cullin-F box protein (SCF) complex or the anaphase-promoting complex (APC). Here we describe an unconventional cell cycle-regulated proteolytic mechanism that acts on the Acm1 protein, an inhibitor of the APC activator Cdh1 in budding yeast. Although Acm1 can be recognized as a substrate by the Cdc20-activated APC (APCCdc20) in anaphase, APCCdc20 is neither necessary nor sufficient for complete Acm1 degradation at the end of mitosis. An APC-independent, but 26S proteasome-dependent, mechanism is sufficient for complete Acm1 clearance from late mitotic and G1 cells. Plant WEE1 kinase is cell cycle regulated and removed at mitosis via the 26S proteasome machinery.This article is from Journal of Experimental Botany , volume 64 . Abstract In yeasts and animals, premature entry into mitosis is prevented by the inhibitory phosphorylation of cyclin-dependent kinase (CDK) by WEE1 kinase, and, at mitosis, WEE1 protein is removed through the action of the 26S proteasome. Although in higher plants WEE1 function has been confirmed in the DNA replication checkpoint, Arabidopsis wee1 insertion mutants grow normally, and a role for the protein in the G2/M transition during an unperturbed plant cell cycle is yet to be confirmed. Here data are presented showing that the inhibitory effect of WEE1 on CDK activity in tobacco BY-2 cell cultures is cell cycle regulated independently of the DNA replication checkpoint: it is high during S-phase but drops as cells traverse G2 and enter mitosis. DTIC ADA357346: Biology of Somatostatin and Somatostatin Receptors in Breast Cancer.Fifty additional primary human breast tumor samples were analysed for SSTR1-5 mRNA and shown to be all receptor positive. Mean SSTR mRNA level was the highest for SSTR3 followed by SSTR1, SSTR2, SSTR5, and SSTR4. Statistical analysis showed a negative correlation between SSTR2 and tumor size, a positive correlation between SSTR2 and estrogen receptors (ER), and a trend towards a positive correlation between SSTR2 and progesterone receptor, SSTR1 and ER, SSTR1 and lymph node status, and SSTR3 and tumor grade. Breast cancer cell lines also expressed multiple SSTR subtypes but the level of receptor expression was less than that in solid tumors. A panel of antibodies against hSSTR1-5 was produced which detected all five SSTR antigens by Western blots and immunocytochemistry. Aberrations in the ubiquitin-mediated destruction pathways, and particularly in the destruction of the Dl cyclin, have been implicated in human breast cancer. A cascade of enzymes leads to ubiquitination of substrate: El enzyme (ubiquitin activating); E2 enzyme (ubiquitin conjugating), and E3 enzyme (ubiquitin ligase). This last class of enzyme is large, and it performs the functions of targeting substrate and catalyzing formation of the polvubiquitin chains that mark substrate for destruction. Functional inactivation or reduction of this pathway is a common event found in many types of human tumors. To test how the Rb pathway interacts with other cellular pathways in tumor suppression, we characterized mice with combined mutations in the CDK inhibitor p18 (exp Ink4c) and the lipid phosphatase PTEN, which regulates cell growth and survival.
