| DTIC ADA453395: Suppression of Prostate Cancer by PTEN and p18INK4c | The Rb pathway suppresses tumorigenesis by constraining G1 cell cycle progression. Functional inactivation or reduction of this pathway is a common event found in many types of human tumors. To test how the Rb pathway interacts with other cellular pathways in tumor suppression, we characterized mice with combined mutations in the CDK inhibitor p18 (exp Ink4c) and the lipid phosphatase PTEN, which regulates cell growth and survival. The p18(exp -/-) ; PTEN (exp +/-) double mutant mice develop prostate cancer in the anterior and dorsolateral lobes and thyroid C-cell tumors with nearly complete penetrance, and pituitary tumors in both the anterior and intermediate lobes. Aberrations in the ubiquitin-mediated destruction pathways, and particularly in the destruction of the Dl cyclin, have been implicated in human breast cancer. A cascade of enzymes leads to ubiquitination of substrate: El enzyme (ubiquitin activating); E2 enzyme (ubiquitin conjugating), and E3 enzyme (ubiquitin ligase). | DTIC ADA331989: Modulation of Cyclin Expression by c-Myc in Malignant and Nonmalignant Mammary Epithelial Cells. | The purpose of this grant was to support predoctoral training on an experimental model relevant to breast cancer etiology. Specifically, we explored the multifactorial nature of the interaction a growth factor (TGFalpha, which is commonly expressed both in benign and malignant proliferative disease of the human breast) and an oncogene (c-myc, whose gene is amplified and whose protein is inappropriately expressed in 20-30% of human breast tumors. Using a transgenic mouse model we observed that co-expression of these two genes was remarkably synergistic for onset and progression of cell survival and proliferation. While mye induced both p53 and bax death-promoting genes, TCFalpha promoted cell survival by inducing Bc1XL. In terms of the cell cycle, mye shortened G1 by modulating cyc1lin E, p27, and cdc25A, which activated cdk-2 and inactivated Rb. Abstract The administration of exogenous β-hydroxybutyrate (β-HB), as well as fasting and caloric restriction, is a condition associated with β-HB abundance and decreased appetite in animals. Increased β-HB and decreased appetite exist simultaneously in some diseases, such as bovine left displaced abomasums (LDA) and human chronic gastritis. However, the effects of β-HB on stomach injuries have not been explored. To elucidate the possible effects of exogenous β-HB on the stomach, mice were injected intraperitoneally with β-HB, and bovine abomasum smooth muscle cells (BSMCs) were treated with different concentrations of β-HB.
